comment on [PSA] your bac water is fine, your fridge door is not in c/reconstitution · 2 points · 1 days ago

a U-100 syringe reads 100 units to the millilitre, every time

comment on [Question] does needle dead space actually matter at 2.5mg doses in c/reconstitution · 11 points · 2 days ago

Shook the first vial because nobody told me not to.

mateusz_achebe has the units point exactly right. They are a volume on the barrel and nothing else.

comment on [Method] the gradient everyone uses for sema and why it under-resolves the des-amido in c/hplc · 151 points · 4 days ago

Carryover from a previous high-concentration injection looks exactly like a small impurity. The blank injection is four minutes and it removes the ambiguity entirely.

comment on [Long Term] found my lowest effective dose by going down, not up in c/maintenancephase · 0 points · 11 days ago

Small fix — you have described interval stretching and called it a dose reduction. The exposure profiles differ.

permalink

comment on [Discussion] non-peptide agonism is a genuinely different engineering problem in c/orforglipron · 12 points · 13 days ago

Push back: daily dosing is not automatically better adherence. It is a different failure mode, not a solved problem.

comment on [Results] 79 weeks, 21kg, and non-peptide was the hard part in c/orforglipron · 1 points · 23 days ago

Cosigning on daily dosing. It changes the exposure profile and it changes adherence, in both directions.

permalink

comment on [Long Term] what my labs look like three years in in c/maintenancephase · 1 points · 28 days ago

stretching the interval is not the same as reducing the dose

permalink

comment on help me understand non-peptide, I have read the wiki twice in c/orforglipron · 22 points · 1 months ago

daily dosing changes adherence in both directions

permalink

comment on how much of what we believe about mg/mL actually comes from reconstitution threads in c/reconstitution · 176 points · 1 months ago

Correction: that gives 40 units, not what you have written. 5mg into 2mL is 2.5mg/mL and the division follows from there.

comment on [Question] do you tell new people how long this actually is in c/maintenancephase · 4 points · 1 months ago

What is the plan if the drift continues for another month?

permalink

comment on genuine question about purity that I am slightly embarrassed to ask in c/hplc · 50 points · 1 months ago

I would not blame the sample yet. Everything you have described is consistent with the column rather than the vial.

comment on someone explain concentration to me like I have not read a paper in years in c/reconstitution · 17 points · 1 months ago

label the vial with the date as well as the concentration

Carryover from a previous high-concentration injection looks exactly like a small impurity. The blank injection is four minutes and it removes the ambiguity entirely.

comment on [Discussion] insulin syringe is doing more work than we give it credit for in c/reconstitution · 218 points · 1 months ago

Bacteriostatic water contains benzyl alcohol as a preservative, which is what makes repeated entry into the same vial reasonable. Plain sterile water has no preservative.

permalink

comment on [Discussion] the bacteriostatic water advice in here is 3 years out of date in c/reconstitution · 121 points · 1 months ago

The lyophilised cake displaces a small volume, so the true concentration is very slightly lower than the arithmetic suggests. It is a fraction of a percent and it is dwarfed by fill tolerance.

comment on regain: what the trials say vs what this community says in c/maintenancephase · 130 points · 1 months ago

Dose reduction and interval extension are different interventions. Halving the dose keeps the rhythm and lowers the level; stretching keeps the level and lowers the frequency, with a lower trough.

comment on switched from 10mg to 15mg and constipation got better, not worse in c/orforglipron · 1 points · 2 months ago

Correction: that is the oral peptide product, which is a different thing entirely. This one is a small molecule.

permalink

comment on am I the only one who found long term harder than the injections in c/maintenancephase · 31 points · 3 months ago

Not convinced. Interval stretching changes your exposure profile, not just the total, and the two are not equivalent.

permalink

comment on the long term thing finally clicked for me and I want to write it down in c/maintenancephase · 1 points · 3 months ago

What dose are you holding, and for how long now?

permalink

comment on the long term thing finally clicked for me and I want to write it down in c/maintenancephase · 37 points · 3 months ago

Withdrawal studies in this class consistently show substantial regain after discontinuation. That is a pharmacological finding about a chronic condition, not a statement about anyone’s discipline.

comment on [Meta] proposal — a flair for long term posts in c/maintenancephase · 41 points · 3 months ago

the goal weight you picked at week 1 was a guess

Disagreeing with this bit: a two-kilo range is not drift and treating it as a problem creates one.

permalink

comment on [Results] 55 weeks on 5mg, full numbers, ask me anything boring in c/orforglipron · 17 points · 3 months ago

Analytically this is a small-molecule identity and purity problem, not a peptide one. The methods, the impurity classes and the reference standards are all different.

Has appetite changed, or only the scale?

Drifted up 21kg over a winter and panicked. It came back off without any change, which is apparently just what weight does.

KP sent me the column, the gradient and the theoretical mass without being asked. That is a short list and they are on it.

Disagreeing with this bit: a two-kilo range is not drift and treating it as a problem creates one.

Not convinced by that integration. Dropping the baseline there absorbs part of the shoulder into the main peak.

Same. Monthly is plenty at this stage and daily weighing made me miserable for no information gain.

Yes — nothing containing this is approved anywhere, and the threads keep forgetting it.
