comment on FLOW got published and nobody in here is talking about eGFR slope in c/trialwatch · 1 points · 2 days ago

This. Intention-to-treat versus completer analysis routinely moves the headline by several points.

comment on what does titration actually mean in c/newbiequestions · 0 points · 3 days ago

Concentration is mass over volume, and a U-100 syringe reads 100 units per millilitre. Those two facts are the whole of the arithmetic this board keeps explaining.

comment on [PeptideMeter] QSC tirz — 97.1%, second sample from the same batch in c/labresults · 5 points · 14 days ago

Inter-lab variance on this assay runs roughly a point either way, driven mostly by gradient and integration. Two results inside that band agree; they do not disagree quietly.

comment on [Paper] ATTAIN-1 — an oral pill with no food restrictions is a bigger deal than the % suggests in c/trialwatch · 57 points · 17 days ago

Read a press release and the publication three months apart. The hedging in the second one was substantial.

titration means going slow on the way up

I confused two very similar words on my first order and asked here, expecting to be mocked. Three people explained it properly instead.

comment on stretching to 10 days instead of 7 — anyone doing this long term in c/maintenancephase · 65 points · 24 days ago

Appetite returning before the scale moves follows from the mechanism: the appetite effect is the proximal one and weight is the lagging integrated outcome.

comment on three years of SURMOUNT threads, summarised so you do not have to read them in c/trialwatch · 14 points · 29 days ago

Open-label extensions lose their randomisation. Anybody still enrolled at week 104 is a selected group and the numbers describe that group.

comment on silly question but which day of the week should i pick in c/newbiequestions · 6 points · 1 months ago

The injection was scarier in my head than in reality. It took about ten seconds and I felt slightly silly afterwards.

comment on [Meta] the FLOW rule is doing its job and people should stop complaining in c/trialwatch · 123 points · 1 months ago

A confidence interval is the range of effects compatible with the data. Two trials with overlapping intervals have not disagreed, whatever their point estimates look like next to each other.

comment on the gallbladder question that gets asked weekly, answered properly in c/sideeffects · 1 points · 1 months ago

Agreed. Slowing down at meals did more for my sulphur burps than everything I bought to fix it, which was embarrassing to discover.

comment on how much of what we believe about heart rate actually comes from sulphur burps threads in c/sideeffects · 1 points · 1 months ago

Reflux was the one that got me. Stopped eating in the three hours before bed and it went from nightly to almost never.

comment on how much of what we believe about heart rate actually comes from sulphur burps threads in c/sideeffects · 1 points · 1 months ago

The fatigue turned out to be that I was eating about a third of what I thought I was. Tracked it for a week and the mystery evaporated.

comment on how much of what we believe about endpoint actually comes from SURPASS threads in c/trialwatch · 17 points · 1 months ago

The discontinuation numbers were the most useful thing in the paper for me and they were in a supplementary table.

Which day of the week relative to the shot does it hit?

Agreed, with the caveat that "usually settles" is not "always settles" and the exceptions should still get asked about properly.

appetite change usually shows up before the scale does

comment on normal is the most under-discussed thing on this board in c/newbiequestions · 5 points · 1 months ago

Research-use-only material is not approved for human use. That is a plain statement of regulatory fact and it belongs in every beginner thread.

Wrote the concentration on the vial after reading it here. Have never once had to reconstruct it from memory since.

permalink

comment on [Discussion] the heart rate advice in here is 2 years out of date in c/sideeffects · 11 points · 2 months ago

I would be careful here. "Everyone gets this" is not true, and it makes the people who get something unusual assume they are fine when they should be asking.

permalink

comment on genuine question about sulphur burps that I am slightly embarrassed to ask in c/sideeffects · 88 points · 2 months ago

fatigue for the first eight days after each step, then nothing, every single time. Predictable enough that I started planning my week around it.

permalink

comment on anyone else notice first time kicking in around week 66 in c/newbiequestions · 1 points · 2 months ago

Careful — that is a dosing recommendation dressed as an anecdote, and this board removes those.

comment on am I the only one who found long term harder than the injections in c/maintenancephase · 7 points · 3 months ago

Wrote down what maintenance would look like at about week 21, before I needed it. Having decided in advance made the transition boring.

comment on am I the only one who found dose stretching harder than the injections in c/maintenancephase · 0 points · 3 months ago

Withdrawal studies in this class consistently show substantial regain after discontinuation. That is a pharmacological finding about a chronic condition, not a statement about anyone’s discipline.

permalink

comment on week 95 check-in — 48kg down, nausea manageable, one thing confusing me in c/maintenancephase · 0 points · 4 months ago

Maintenance is the phase with the least published guidance and the most individual variation, which is precisely why board consensus here is worth less than usual.

permalink

comment on stalled for 2 weeks at 15mg and I refuse to panic this time in c/newbiequestions · 108 points · 4 months ago

Appetite change and weight change are different measurements on different timescales. The first is usually noticed first and the second lags it.

comment on anyone else notice fatigue kicking in around week 46 in c/sideeffects · 9 points · 4 months ago

write down when it starts and stops, the pattern is the useful part
