comment on FLOW got published and nobody in here is talking about eGFR slope in c/trialwatch · 1 points · 2 days ago

Careful with that mean.

Disagreeing with this line: that is a relative reduction and the absolute numbers are considerably less dramatic.

Adding more water does not change how much peptide is in the vial. It changes the concentration, which changes the volume you draw for the same amount. That is the entire relationship.

comment on sterile water is not bacteriostatic water and it is not close in c/reconstitution · 46 points · 5 days ago

Bacteriostatic water contains benzyl alcohol as a preservative, which is what makes repeated entry into the same vial reasonable. Plain sterile water has no preservative.

comment on [Side Effects] alcohol hits completely differently now and nobody mentioned it in c/sideeffects · 12 points · 9 days ago

Correction: that is a known effect of reduced intake rather than a direct drug effect. The distinction matters because the fix is different.

Chased a ghost peak for three weeks. It was the plumbing. It is always the plumbing.

Right, and the hair thing is worth repeating: it tracks the rate of loss rather than the compound, and it stops.

DEXA has a meaningful coefficient of variation, and it differs between machines and protocols. Comparing scans from different clinics adds a systematic offset that is invisible on the report.

comment on [PSA] let the alcohol swab dry before you puncture, that is the step everyone skips in c/reconstitution · 67 points · 15 days ago

The lyophilised cake displaces a small volume, so the true concentration is very slightly lower than the arithmetic suggests. It is a fraction of a percent and it is dwarfed by fill tolerance.

comment on is it bad that i am not nauseous at all in c/newbiequestions · 46 points · 16 days ago

Appetite change and weight change are different measurements on different timescales. The first is usually noticed first and the second lags it.

comment on [Lab] LC-MS confirmed identity but the UV purity is 94%. now what in c/hplc · 276 points · 17 days ago

Area percent is the integrated area of your peak over the total integrated area at one wavelength on one gradient. Change any of those and the number changes without the sample changing.

Research-use-only material is not approved for human use. That is a plain statement of regulatory fact and it belongs in every beginner thread.

comment on watch where the curves separate, not just whether they do in c/trialwatch · 22 points · 22 days ago

Discontinuation rates are a tolerability result. A trial with a strong efficacy number and heavy discontinuation is telling you two things and people only quote one.

comment on someone explain units to me like I have not read a paper in years in c/reconstitution · 1 points · 23 days ago

Agreed — units are volume. Once that lands, the rest of the arithmetic is one division.

comment on [Explainer] why an oral non-peptide escapes the absorption problem entirely in c/glp1science · 167 points · 25 days ago

Receptor expression in a tissue is necessary but not sufficient for an effect. You also need the agonist to reach it at relevant exposure, and that is where a lot of confident mechanism talk falls down.

Correcting myself upthread: I said the gradient was linear and looking again it has a hold in the middle.

Correcting my own post: the payment date was the 14th, not the 4th. The gap is smaller than I made it sound.

Tested three lots over a year rather than three vials from one lot. Cost the same and told me something completely different.

comment on [Results] 78 weeks on 7.5mg, full numbers, ask me anything boring in c/glp1muscle · 46 points · 1 months ago

Reduced appetite makes hitting a protein target a logistics problem rather than a motivation problem, which is why the practical advice here is about timing and food choice rather than willpower.

That is sterile water, not bacteriostatic. Same appearance, different product, and it matters for a multi-draw vial.

comment on [Question] HPLC — what am I missing here in c/hplc · 205 points · 1 months ago

UV response is not uniform across species. At 214nm you are looking at the peptide bond, which is why it is the working wavelength for this class; at 280nm you only see tryptophan, tyrosine and phenylalanine.

If the report gives a mass spectrum as well as UV purity, say so. Identity and relative quantity are separate claims and the log tracks both.
