Overview — page 3 of 3

comment on three years of reflux threads, summarised so you do not have to read them in c/sideeffects · 1 points · 1 year ago

That reads like advice, and none of us can give it. Describe what you did, not what somebody else should do.

comment on [Results] 37 weeks, 6kg, and weekly dosing was the hard part in c/semaglutide · 77 points · 1 year ago

The escalation schedule in the trials existed to keep people on the drug. It is a tolerability ramp, and it is why the label reads the way it does.

comment on unpopular opinion: most of what gets said here about goal weight is guesswork in c/maintenancephase · 16 points · 1 year ago

Right — appetite returns before the scale moves, which is the early signal people miss.

permalink

comment on anyone else notice basics kicking in around week 49 in c/newbiequestions · 91 points · 1 year ago

research-use-only material is not approved for human use, and that belongs in every beginner thread

Did you write the concentration on the vial?

permalink

comment on the Wegovy question that gets asked weekly, answered properly in c/semaglutide · 122 points · 1 year ago

do not chase somebody else’s titration schedule, they are not you

comment on switched from 1.7mg to 2.4mg and nausea got better, not worse in c/maintenancephase · 1 points · 1 year ago

Push back: that is not a plateau, that is maintenance. You have arrived and you are describing it as a failure.

permalink

comment on am I the only one who found plateau harder than the injections in c/semaglutide · 101 points · 1 year ago

Not convinced. You changed the dose and the week at the same time, so you cannot attribute the change to either.

comment on [Question] how do you actually verify half-life in c/glp1science · 423 points · 1 year ago

the peripheral and central stories are not in competition

Disagreeing with this specific inference — that is a preclinical result being read as human pharmacology.

permalink

comment on 15 months in and struggle is still the thing I get wrong in c/progresspics · 28 points · 1 year ago

Correction: 17kg over 12 months is a different claim from 17kg in a month, and the post reads as the second.

comment on why does nobody talk about appetite in c/glp1science · 56 points · 1 year ago

Are we talking about receptor affinity or clinical potency?

permalink

comment on why does nobody talk about appetite in c/glp1science · 140 points · 1 year ago

That study was in a rodent model. Worth stating, since the thread has been reading it as human data.

permalink

comment on [Meta] the Wegovy rule is doing its job and people should stop complaining in c/semaglutide · 148 points · 1 year ago

Correct, and worth adding: the trial average is an average. Half the people in it did worse than 15% and were still on the drug.

comment on anyone else notice hypoglycaemia kicking in around week 8 in c/t2dglp1 · 1 points · 1 year ago

dose for glycaemic control is not the same conversation as dose for weight

permalink

comment on anyone else notice hypoglycaemia kicking in around week 8 in c/t2dglp1 · 5 points · 1 year ago

Small fix — insulin secretion in this class is glucose-dependent, which is precisely why the risk you describe comes from the other agent.

comment on reflux — 5 things I got wrong before I got it right in c/sideeffects · 119 points · 1 year ago

Right, and the hair thing is worth repeating: it tracks the rate of loss rather than the compound, and it stops.

comment on [Results] 12 weeks on 1.7mg, full numbers, ask me anything boring in c/progresspics · 17 points · 1 year ago

waist is the measurement that keeps moving during a scale stall

permalink

comment on why does nobody talk about regain in c/maintenancephase · 1 points · 1 year ago

stretching the interval is not the same as reducing the dose

comment on [PSA] TB-500 is not what most of this community thinks it is in c/researchpeptides · 4 points · 1 year ago

Purity methods differ by compound class. A gradient and wavelength developed for one peptide will not necessarily resolve the relevant impurities of another.

comment on goal weight — my 23-week log, condensed into one table in c/maintenancephase · 38 points · 1 year ago

Same. Monthly is plenty at this stage and daily weighing made me miserable for no information gain.

permalink

comment on week 59 check-in — 17kg down, early fullness manageable, one thing confusing me in c/sideeffects · 0 points · 1 year ago

Has anything else changed in the same window?

permalink

comment on someone explain receptor to me like I have not read a paper in years in c/glp1science · 0 points · 1 year ago

Small fix — it slows gastric emptying, it does not halt it, and the distinction matters for the mechanism you are proposing.

permalink

comment on why does nobody talk about maintenance in c/maintenancephase · 6 points · 1 year ago

Wrote down what maintenance would look like at about week 78, before I needed it. Having decided in advance made the transition boring.

permalink

comment on thymosin — 16 things I got wrong before I got it right in c/researchpeptides · 1 points · 1 year ago

That method was developed for a different compound class and would not resolve the impurities you are asking about.

permalink

comment on [Results] 58 weeks, 21kg, and plateau was the hard part in c/semaglutide · 1 points · 1 year ago

Minor fix — the half-life is about a week, not about a day.

Right, and the timescale is the bit that keeps getting lost. Judge a step at a month, not at a weekend.

permalink

comment on genuine question about nausea that I am slightly embarrassed to ask in c/sideeffects · 42 points · 1 year ago

Disagree. Severe abdominal pain is not on the list of things to wait out, and this board should not be treating it as one.

permalink

comment on am I the only one who found struggle harder than the injections in c/progresspics · 23 points · 1 year ago

Agreed that goal weights picked at the start are guesses. Mine was, and I revised it twice.

permalink

comment on [Discussion] the long term advice in here is 3 years out of date in c/maintenancephase · 290 points · 1 year ago

That reads like a taper schedule and this board cannot hand those out.

permalink

comment on [Discussion] the A1c advice in here is 2 years out of date in c/t2dglp1 · 9 points · 1 year ago

The diabetes and obesity programmes are separate, with different populations and different primary endpoints. Reading a result across from one to the other is not a comparison.

Is the goal you set at the start still the goal?

permalink

comment on [Meta] the goal rule is doing its job and people should stop complaining in c/progresspics · 49 points · 2 years ago

I would not treat a two-kilo range as a regain. Weight varies day to day in everybody.

comment on my TSH moved and I cannot work out whether sulphur burps are why in c/sideeffects · 52 points · 2 years ago

Same pattern here.

step_therapy_s is right about the timing. The day-of-week pattern is the diagnostic bit, not the severity.

comment on [Vendor] QYB vs HJ on the same compound, same month — both cleared 99.0% in c/semaglutide · 1 points · 2 years ago

This matches me almost exactly, right down to the early fullness showing up on the second day and being gone by the fourth.

permalink

comment on [Results] 45 weeks, 14kg, and beginner was the hard part in c/newbiequestions · 2 points · 2 years ago

Yes. The starting dose is a tolerance step. It is not supposed to do very much.

permalink

comment on [Discussion] the nausea advice in here is 3 years out of date in c/sideeffects · 28 points · 2 years ago

Nothing in this thread is medical advice, including this comment. I am describing a mechanism, not telling anybody what to do about their own week.

comment on what changed for me between month 8 and month 12 in c/sideeffects · 512 points · 2 years ago

The fatigue turned out to be that I was eating about a third of what I thought I was. Tracked it for a week and the mystery evaporated.

permalink

Body weight varies by a couple of kilos over days in stable people through fluid, glycogen and gut content. A range is not a trend.

permalink

comment on someone explain incretin to me like I have not read a paper in years in c/glp1science · 45 points · 2 years ago

The half-life explanation was the thing that made weekly dosing intuitive for me rather than arbitrary.
