comment on [Question] how do you actually verify sequence in c/bpc157 · 213 points · 1 year ago

The published literature is overwhelmingly preclinical, largely in rodent models. Translating a rodent result into a human expectation is not a small step, and almost every confident claim here takes it silently.

Fertility changed faster than I expected with the weight loss. Nobody had raised it and I wish somebody had.

Independent purity testing on this compound is thin compared with the older molecules, simply because fewer members have paid for it. Fewer results means wider uncertainty, not a verdict.

hair shedding follows the weight loss, not the drug, and it stops

A certificate with a purity figure and no method line is not falsifiable. You cannot reproduce it, you cannot check it, and it tells you nothing about how the number was produced.

comment on why does nobody talk about Zepbound in c/tirzepatide · 1k points · 1 year ago

Weekly dosing again, half-life in the same neighbourhood as sema, so a step change takes about a month to reach steady state. Judging a new step at day five is judging noise.

kavya_kravchenko is right about the timing.

Adding to this — holding the dose rather than stepping is the option people forget they have.

Has anyone posted an independent result on that batch?

Agreed that the vocabulary here is imprecise and it causes people to answer different questions to the ones asked.

comment on the nausea thing finally clicked for me and I want to write it down in c/sideeffects · 353 points · 1 year ago

A symptom that begins the day after the shot and fades by day four tracks the exposure curve. One that appears at random in week 57 probably does not, and that is worth saying to a clinician rather than to us.

ask for the mass spectrum, not just the purity number

Is that a clinical claim or a preclinical one?

muscle cramps for the first eight days after each step, then nothing, every single time. Predictable enough that I started planning my week around it.

the muscle cramps profile is genuinely gentler than people expect coming from sema

Yes — the comparison threads are apples to oranges and they generate more heat than anything else here.

comment on unpopular opinion: most of what gets said here about phase 2 is guesswork in c/retatrutide · 386 points · 1 year ago

Glucagon receptor agonism is associated with increased energy expenditure and with hepatic effects. That is a mechanistic story with strong preclinical support and limited phase 2 human data.

Appetite effect for me is flat across seven days. On sema it was a wave. Same person, different molecule.

Right, and the hair thing is worth repeating: it tracks the rate of loss rather than the compound, and it stops.

comment on [Results] 57 weeks, 25kg, and glucagon was the hard part in c/retatrutide · 526 points · 1 year ago

Nothing containing this compound is approved anywhere. Research-use-only material is not approved for human use, and that is a statement of fact rather than a disclaimer.

Supply notices are issued nationally. A notice in one member state does not imply availability problems in another, even for the same product.

The thing that decided it for me was that MKM answered a column-and-gradient question in writing without asking why I wanted to know.

Right, and a shortage notice in one country tells you very little about the next one along.

permalink

comment on reading triple agonist threads from 2024 and half of it aged badly in c/retatrutide · 329 points · 2 years ago

It is a triple agonist at the GLP-1, GIP and glucagon receptors. The glucagon arm is the genuinely novel component and it is the one with the least human outcome data behind it.

Hunan GoodPeptides Biotech came up in three threads the week I was ordering and I could not find a single independent result for them anywhere.

Adding the practical version: ask for lead time, minimum quantity and the batch record in the same message. Three answers, one email.

Constipation is mostly reduced intake plus reduced fluid, not a direct effect. That is why the boring fibre and water answer keeps working.
