comment on [Stats] relative risk reduction sells papers, absolute risk reduction makes decisions in c/trialwatch · 71 points · 2 days ago
Intention-to-treat analyses everybody randomised regardless of what they did afterwards. Completer analyses only those who finished. The second is systematically more flattering and both are legitimate if labelled.
comment on [Question] does needle dead space actually matter at 2.5mg doses in c/reconstitution · 11 points · 3 days ago
What is written on the vial right now?
comment on [Trial Data] STEP 1 vs SURMOUNT-1 side by side, with the caveats in c/trialwatch · 31 points · 5 days ago
Open-label extensions lose their randomisation. Anybody still enrolled at week 104 is a selected group and the numbers describe that group.
comment on the food noise going quiet is the part nobody warned me about in c/semaglutide · 187 points · 8 days ago
Weight change and appetite change are different endpoints on different timescales. Conflating them is where most of the confusion on this board starts.
comment on cloudy after reconstitution — discard, right? right. in c/reconstitution · 67 points · 13 days ago
The dead space question sent me down a rabbit hole for a week. It is a fraction of a unit and it is the same every draw.
comment on [PSA] let the alcohol swab dry before you puncture, that is the step everyone skips in c/reconstitution · 17 points · 14 days ago
units are a volume on a U-100 syringe, not a dose
comment on help me understand insulin syringe, I have read the wiki twice in c/reconstitution · 3 points · 18 days ago
20mg into 2mL is 10mg/mL, and 0.5mg of that is 5 units
Adding one thing — put the date on the label too. Concentration plus date and you can never mix two vials up.
comment on [Titration] stayed at 0.5 for eleven weeks and it was the right call in c/semaglutide · 54 points · 19 days ago
Steady state after a dose change takes four to five half-lives, so the fair judgement point on a new step is about a month, not four days.
comment on does anyone else get one good week and one flat week on repeat in c/semaglutide · 6 points · 1 months ago
I would push back gently. Going up because the scale paused is the single most common mistake described on this board.
comment on went back down from 2.4 to 1.7 and lost more. explain that in c/semaglutide · 0 points · 1 months ago
Research-use-only material is not approved for human use, so anything in this thread about how a compounded or research vial "should" behave is a chemistry discussion, not a dosing one.
comment on STEP 1 was -14.9% average and i keep having to remind myself average means average in c/semaglutide · 23 points · 1 months ago
Same. I sat at 1.7mg for three months because it was working and there was nothing to fix.
comment on what would you tell week-1 you about 2.4mg in c/semaglutide · 32 points · 2 months ago
Disagree. 2.4 is the top of the label, not the goal, and "everyone ends up there" is just survivorship in the posts you read.
comment on the titration question that gets asked weekly, answered properly in c/semaglutide · 217 points · 2 months ago
Careful. A four-week stall is inside normal variation and treating it as failure is how people escalate for no reason.
comment on [Discussion] TRIUMPH is doing more work than we give it credit for in c/trialwatch · 22 points · 2 months ago
a press release is not a publication
Agreed. And the interval, not the point estimate, is what the trial actually established.
comment on how much of what we believe about endpoint actually comes from FLOW threads in c/trialwatch · 20 points · 2 months ago
Correction: SURMOUNT is the obesity programme and SURPASS is the diabetes one. The figure you quoted belongs to the other one.
comment on [PSA] 2.4mg is not what most of this community thinks it is in c/semaglutide · 89 points · 3 months ago
dose day drift of a day either way is fine, the curve barely notices
comment on unpopular opinion: most of what gets said here about bacteriostatic water is guesswork in c/reconstitution · 14 points · 3 months ago
Yes, and the dead-space point is worth repeating: it is small, it is real, and it is the same for every draw.

comment on [Question] how do you actually verify Ozempic in c/semaglutide · 143 points · 5 months ago
That reads like a titration plan and this board cannot give you one. Somebody who actually knows your history has to.
comment on someone explain FLOW to me like I have not read a paper in years in c/trialwatch · 31 points · 6 months ago
Do you have the publication or the press release?
comment on mg/mL — 16 things I got wrong before I got it right in c/reconstitution · 14 points · 6 months ago
Bacteriostatic water contains benzyl alcohol as a preservative, which is what makes repeated entry into the same vial reasonable. Plain sterile water has no preservative.
comment on [Meta] the FLOW rule is doing its job and people should stop complaining in c/trialwatch · 75 points · 6 months ago
A confidence interval is the range of effects compatible with the data. Two trials with overlapping intervals have not disagreed, whatever their point estimates look like next to each other.

25kg over 6 months and honestly the most useful marker was that stairs stopped being a decision.

Agreed that goal weights picked at the start are guesses. Mine was, and I revised it twice.

How many mg in the vial and how many mL are you adding?
comment on [Discussion] TRIUMPH is doing more work than we give it credit for in c/trialwatch · 6 points · 9 months ago
Yes. The appendix tables are where the subgroup and the adverse event detail actually live.

Asked WWB what volume they reconstitute to for their own stability testing and they answered in one line.
ilias_cabrera has the units point exactly right. They are a volume on the barrel and nothing else.

Cosigning the round-number approach. Reconstituting to something awkward is how you end up drawing 27.5 units at six in the morning.

comment on [Question] how do you actually verify SURPASS in c/trialwatch · 382 points · 1 year ago
Discontinuation rates are a tolerability result. A trial with a strong efficacy number and heavy discontinuation is telling you two things and people only quote one.

comment on does reconstitution actually matter or is it forum lore at this point in c/reconstitution · 63 points · 1 year ago
That is sterile water, not bacteriostatic. Same appearance, different product, and it matters for a multi-draw vial.
