comment on [Method] the gradient everyone uses for sema and why it under-resolves the des-amido in c/hplc · -38 points · 3 days ago
A shoulder that does not baseline-resolve cannot be quantified honestly. You can report it as an unresolved shoulder, which is useful information, or you can develop the method until it resolves.
comment on peak at 4.2 min, nobody can tell me what it is, ideas in c/hplc · 10 points · 5 days ago
Same method at both labs, or two different gradients?
comment on week two, nothing is happening, everyone says that is normal, is it in c/newbiequestions · 17 points · 5 days ago

comment on what does 0.25 mean, 0.25 of what in c/newbiequestions · 1 points · 13 days ago
I would not panic about that. It is on the list of ordinary first-month experiences.
comment on [Titration] 12 weeks per step is not too slow, it is just unfashionable in c/tirzepatide · 164 points · 14 days ago
This matches mine. Milder muscle cramps than I expected, and what there was settled inside a fortnight of each step.
comment on what does 0.25 mean, 0.25 of what in c/newbiequestions · 11 points · 15 days ago
Small correction, said kindly: those are two different products with similar names, and the difference matters for a multi-draw vial.
comment on SURMOUNT-1 was -20.9% at 72 weeks. that is the number, not -30% in c/tirzepatide · 32 points · 16 days ago
2.5mg is a four-week starting dose, not a maintenance dose. Worth being precise since people search these threads.
comment on [Caution] not approved anywhere. keep posts descriptive. in c/cagrilintide · 1 points · 20 days ago
The engineering problem was duration: native amylin is short-acting and aggregation-prone.
Adding the standing caveat — nothing here is approved standalone and research material is not for human use.
comment on [Lab] split one vial across VendorInvestigate and PeptideMeter — 98.0% and 97.9% in c/hplc · 9 points · 26 days ago
Started reporting my own integrations with the baseline choice stated. Arguments in my threads dropped by about half.
comment on why do people talk about units and mg like they are different things in c/newbiequestions · 22 points · 1 months ago
Bacteriostatic water contains a preservative, which is what makes repeated entry into a vial reasonable. Sterile water does not, and the two are not interchangeable for that purpose.
comment on [Discussion] we are measuring half-life at the wrong time and calling it noise in c/glp1science · 23 points · 1 months ago
a mechanism you can state is not a mechanism you have demonstrated
comment on genuine question about purity that I am slightly embarrassed to ask in c/hplc · 63 points · 1 months ago
Area percent is the integrated area of your peak over the total integrated area at one wavelength on one gradient. Change any of those and the number changes without the sample changing.
comment on anyone else notice tirzepatide kicking in around week 13 in c/tirzepatide · 140 points · 1 months ago
SURMOUNT-1 ran 72 weeks with the top arm around 21% mean body weight change. SURPASS is the diabetes programme and reports glycaemic endpoints, so quoting the two interchangeably is a category error.
comment on [PSA] HPLC is not what most of this community thinks it is in c/hplc · -20 points · 1 months ago
area percent is relative to what the detector saw and nothing else
comment on what would you tell week-1 you about GIP in c/tirzepatide · 25 points · 1 months ago
Went 10 to 12.5 on the calendar and had the worst fortnight of the whole run. Dropped back to 10 and everything settled.
comment on [Lab] split one vial across Medutest and Janoshik — 99.3% and 98.7% in c/tirzepatide · 0 points · 2 months ago
the reflux profile is genuinely gentler than people expect coming from sema
Not sure about this bit. The GIP arm being real does not tell you that it is what caused your particular week.
comment on incretin: what the trials say vs what this community says in c/glp1science · 41 points · 2 months ago
Receptor expression in a tissue is necessary but not sufficient for an effect.
Agreed — and it is why the central and peripheral stories are complementary rather than rival.

comment on anyone else notice what is kicking in around week 47 in c/newbiequestions · 19 points · 3 months ago
Correction: the starting dose is a tolerance step rather than a treatment dose. The distinction is the answer to the question above.
comment on [Question] how do you actually verify Janoshik in c/labresults · 456 points · 3 months ago
Area percent and net peptide content are different measurements and most reports give one. Quote which, or the comparison downstream is meaningless.
comment on [Lab] 4th independent test on JEEP — 97.3% on a claimed 97.0%, and the trend is the interesting part in c/tirzepatide · 13 points · 4 months ago
Yes — 10mg being a genuine landing place for a lot of people is underrated. There is no medal for 15.
comment on [PSA] incretin is not what most of this community thinks it is in c/glp1science · 14 points · 4 months ago
This. Albumin binding and modification are why the half-life is what it is, and it is a design decision rather than an accident.
comment on why does nobody talk about HPLC in c/hplc · 16 points · 4 months ago
Push back: a longer run is not automatically better resolution. You are trading peak width for time and the ratio is what matters.
comment on [Meta] the mechanism rule is doing its job and people should stop complaining in c/glp1science · 1 points · 4 months ago
GIP receptor biology is genuinely unsettled — there is a live argument about agonism versus antagonism at the receptor — and the clinical results are robust regardless, which is an uncomfortable and interesting position.

comment on [Discussion] SURPASS is doing more work than we give it credit for in c/tirzepatide · -11 points · 5 months ago
Which week did the appetite change actually land for you?
comment on stalled for 4 weeks at 2.5mg and I refuse to panic this time in c/cagrilintide · 33 points · 5 months ago
Not convinced. Amylin signalling is not a GLP-1 pathway and the mechanism you are proposing conflates them.
comment on stalled for 4 weeks at 2.5mg and I refuse to panic this time in c/cagrilintide · 6 points · 5 months ago
Amylin is co-secreted with insulin and acts on satiety and gastric emptying through its own receptor complexes. An amylin analogue is therefore not a variant of an incretin agonist — it is a different signalling axis.

comment on help me understand LC-MS, I have read the wiki twice in c/hplc · 256 points · 6 months ago
That is area percent, not mass percent. The trace cannot give you the second one.
comment on Medutest — 16 things I got wrong before I got it right in c/labresults · 87 points · 7 months ago
net peptide content if the report gives it, and say which you are quoting
comment on reading pharmacology threads from 2024 and half of it aged badly in c/glp1science · 513 points · 7 months ago
GLP-1 receptor agonism acts both peripherally — insulin secretion in a glucose-dependent way, slowed gastric emptying — and centrally, on appetite regulation. The central component is the better explanation for sustained intake reduction.
comment on [Discussion] we are measuring CagriSema at the wrong time and calling it noise in c/cagrilintide · 4 points · 8 months ago
Complementary mechanisms are the rationale for pairing: satiety signalling alongside incretin signalling, rather than more agonism at the same receptor.
comment on three years of 15mg threads, summarised so you do not have to read them in c/tirzepatide · 132 points · 8 months ago
dual agonist, so the GIP arm is doing something the sema threads will not tell you about
comment on [Meta] proposal — a flair for LC-MS posts in c/hplc · 98 points · 11 months ago
Yes — system suitability first. Without it the number is an assertion about the instrument, not the sample.
comment on the GIP question that gets asked weekly, answered properly in c/tirzepatide · 3 points · 11 months ago
It is a dual GIP and GLP-1 receptor agonist, and the GIP arm is the part that has no equivalent in the semaglutide threads. That is why the side-effect profile reads differently rather than just milder.
comment on switched from 1.0mg to 2.4mg and sulphur burps got better, not worse in c/newbiequestions · 103 points · 1 year ago
Gently disagree with the framing — nothing has gone wrong. Week 16 looking flat is the ordinary experience.
