comment on [Question] TRIUMPH — what am I missing here in c/retatrutide · 652 points · 9 months ago

Phase 2 estimates effect and finds a dose range. Phase 3 estimates it precisely in a bigger population and catches what phase 2 was too small to see. Treating them as the same tier of evidence is the recurring error here.

comment on [Discussion] can we stop arguing about appetite until somebody posts a number in c/glp1science · 24 points · 9 months ago

Tolerance to the gastric effect develops with continued exposure while the appetite effect largely persists. That single fact explains most of the "it settles but it still works" pattern the side-effect board reports.

Gave the same trace to two people I trust and got two integrations about a point apart. That was the most educational afternoon I have had here.

Correction: that is glucose-dependent insulin secretion, which is why hypoglycaemia risk is low as monotherapy. Not the same claim as you made.

Same view. Tolerance developing to the gastric effect while the appetite effect persists explains most of what the side-effect board reports.

Mass spectrometry answers identity. UV purity answers relative quantity under the run conditions. A document with one and not the other is answering half the question, and the half it answers should be stated.

Careful — you have a plausible mechanism and no evidence that it is the operative one in the case you are describing.

Retention time shifted 0.4 minutes and I assumed the worst. It was the column temperature.

Cosigning the wavelength point. Half the disagreements in this board are two people looking at different detectors.

Is that a U-100 syringe? The barrel markings change the answer completely.

comment on the appetite question that gets asked weekly, answered properly in c/glp1science · -7 points · 1 year ago

The half-life explanation was the thing that made weekly dosing intuitive for me rather than arbitrary.

Same view — reporting the gradient is what makes a result checkable rather than merely stated.

Asked KP what volume they reconstitute to for their own stability testing and they answered in one line. Useful thing to know.

Bacteriostatic or plain sterile water?

Spent 10 weeks confused about this until somebody drew the table for me. 10mg, 2mL, 5mg/mL, 0.5mg is 10 units. That is the whole thing.

Area percent is the integrated area of your peak over the total integrated area at one wavelength on one gradient. Change any of those and the number changes without the sample changing.

This matches me almost exactly, right down to the muscle cramps showing up on the second day and being gone by the fourth.

comment on am I the only one who found triple agonist harder than the injections in c/retatrutide · 152 points · 1 year ago

Not convinced. You are attributing a week of muscle cramps to the glucagon arm when the escalation rate alone would explain it.

comment on [Question] reconstitution — what am I missing here in c/reconstitution · 0 points · 1 year ago

Not quite.

Right, and the arithmetic behind it: 15mg in 3mL is 5mg/mL, so 0.5mg is 10 units.

comment on help me understand dead space, I have read the wiki twice in c/reconstitution · 20 points · 1 year ago

Reconstituted at 3mL when I meant to use a different volume. Caught it because the units looked wrong on the barrel. Now I write 6.67mg/mL on the glass before I put the cap back.

Small fix — 214nm, not 210. It matters for the comparison you are making with the other run.

Push back: a receptor being expressed in a tissue does not tell you the agonist reaches it at therapeutic exposure.
