comment on [Caution] stacking with a sulfonylurea is a real hypo risk, not a theoretical one in c/t2dglp1 · 15 points · 3 days ago

Yes — the person managing your diabetes is the one to talk to, and this board is explicit about that.

Is this a PCOS diagnosis or an insulin resistance finding?

You cannot infer potency from a temperature record. The record tells you what happened to the box; only a test tells you what happened to the material.

comment on [Paper] amylin co-agonism is genuinely different pharmacology, not just more GLP-1 in c/glp1science · 108 points · 16 days ago

That study was in a rodent model. Worth stating, since the thread has been reading it as human data.

preclinical is not clinical and rodents are not small people

This is the concept everything else on this board is downstream of.

The half-life explanation was the thing that made weekly dosing intuitive for me rather than arbitrary.

comment on [Explainer] the four things a COA must have or it is decorative in c/coa · 95 points · 16 days ago

batch number, date of analysis, method, individual impurities — that is the minimum set

permalink

comment on [COA] full document, nothing cropped, tell me what i am missing in c/coa · 12 points · 1 months ago

Related substances itemised individually is a much stronger document than a single total, because it tells you the shape of the impurity profile rather than its size.

comment on [Results] two years, and the second year mattered more than the first in c/progresspics · 1 points · 1 months ago

Small correction, kindly meant: that is a range rather than a regain. Weight moves day to day in everybody.

comment on reading goal weight threads from 2024 and half of it aged badly in c/maintenancephase · 17 points · 1 months ago

Right — appetite returns before the scale moves, which is the early signal people miss.

That is not what the method line says. It names the technique; it does not certify the operator.

the goal weight you picked at week 1 was a guess

What dose are you holding, and for how long now?

permalink

comment on [PSA] certificate of analysis is not what most of this community thinks it is in c/coa · 19 points · 2 months ago

Pushing back a little: a missing field is usually presentation, not concealment. Ask for the full document before drawing conclusions.

permalink

comment on mechanism is the most under-discussed thing on this board in c/glp1science · 1 points · 3 months ago

GLP-1 receptor agonism acts both peripherally — insulin secretion in a glucose-dependent way, slowed gastric emptying — and centrally, on appetite regulation. The central component is the better explanation for sustained intake reduction.

permalink

comment on [Discussion] we are measuring PCOS at the wrong time and calling it noise in c/glp1women · 124 points · 4 months ago

Push back: PCOS as a diagnosis and insulin resistance as a finding are not interchangeable, and the thread is using them as one.

What are you actually celebrating today? Genuine question.

comment on related substances are the most under-discussed thing on this board in c/coa · 79 points · 5 months ago

Area percent is the peak area of your compound over the total integrated area, at one wavelength, on one gradient. It is a relative measure of what the detector saw, not a mass fraction of the vial.

Read the primary paper after arguing about the summary for a fortnight. The discussion section said almost the opposite of what the thread had concluded.

comment on excursion — 20 things I got wrong before I got it right in c/coldchain · 52 points · 6 months ago

Once in solution, the relevant risks are hydrolysis, aggregation and, for some sequences, oxidation. Refrigeration slows all three; freezing a solution introduces a separate risk at the ice interface.

Agreed on variability. Two people with the same average can have completely different days.

permalink

comment on lowest effective dose — my 7-week log, condensed into one table in c/maintenancephase · 121 points · 6 months ago

Body weight varies by a couple of kilos over days in stable people through fluid, glycogen and gut content. A range is not a trend.

comment on small win: PCOS stopped being a problem at week 61 in c/glp1women · 153 points · 6 months ago

Started logging the cycle in the same sheet as the dose. Three months later the pattern was obvious and it had been invisible before.

Same view. Tolerance developing to the gastric effect while the appetite effect persists explains most of what the side-effect board reports.

My eGFR moved and it turned out to have nothing to do with glucose at all.
