comment on [Vendor] BCH order #16 — Germany, 4 days, no drama, receipts inside in c/coldchain · 2 points · 1 year ago

lyophilised powder is far more forgiving than the panic threads suggest

Agreed. Network first, address second, amount third, and in that order every time.

I would not read that in vitro number across to a person. The conditions are nothing like physiological.

That figure is from the obesity programme, and this thread is about the hepatic one.

Agreed — and the endpoint definitions are where the actual claim lives.

Nothing containing this compound is approved anywhere. Research-use-only material is not approved for human use.

comment on hazard ratio is the most under-discussed thing on this board in c/trialwatch · 20 points · 1 year ago

the confidence interval is the finding, the point estimate is the headline

comment on someone explain cagrilintide to me like I have not read a paper in years in c/cagrilintide · -15 points · 1 year ago

Amylin is co-secreted with insulin and acts on satiety and gastric emptying through its own receptor complexes. An amylin analogue is therefore not a variant of an incretin agonist — it is a different signalling axis.

read the discussion section, that is where the honesty lives

a summer lane and a winter lane are different lanes

write the reconstitution date on the vial

Agreed, and a logger costs less than the vial you are worrying about.

The press release said that; the publication says something more hedged. Worth reading both.

permalink

comment on [Discussion] the excursion advice in here is 2 years out of date in c/coldchain · 71 points · 1 year ago

Careful. "The cold chain failed" is a strong claim and the evidence you have is a warm box.

comment on [Discussion] appetite is doing more work than we give it credit for in c/glp1science · 2 points · 1 year ago

Receptor expression in a tissue is necessary but not sufficient for an effect. You also need the agonist to reach it at relevant exposure, and that is where a lot of confident mechanism talk falls down.

comment on small win: glucagon stopped being a problem at week 36 in c/survodutide · 33 points · 2 years ago

That is an escalation schedule for an unapproved compound and this board does not host those.

permalink

comment on [Question] carrier — what am I missing here in c/intlshipping · 113 points · 2 years ago

Cosigning the seasonality point. February and August on the same lane are different animals.

comment on DHL — 21 things I got wrong before I got it right in c/intlshipping · 459 points · 2 years ago

This. A quoted lead time that holds is worth more than a fast one that does not.

permalink

comment on chargeback — 8 things I got wrong before I got it right in c/cryptopay · -38 points · 2 years ago

Nearly sent on the wrong network. The token symbol was identical and the address format was the only clue.

comment on does half-life actually matter or is it forum lore at this point in c/glp1science · 157 points · 2 years ago

The incretin effect is the observation that oral glucose provokes a larger insulin response than intravenous glucose at matched glycaemia, and the difference is mediated by gut hormones. That is the foundation the whole class sits on.

comment on [Lab] 8th independent test on JEEP — 97.2% on a claimed 97.0%, and the trend is the interesting part in c/survodutide · 14 points · 2 years ago

Cosigning on the glucagon arm. It is the mechanistic thread that connects the hepatic and the metabolic effects.

comment on [PSA] write the concentration on the vial. that is the whole post. in c/trialwatch · 501 points · 2 years ago

A confidence interval is the range of effects compatible with the data. Two trials with overlapping intervals have not disagreed, whatever their point estimates look like next to each other.

Correction: SURMOUNT is the obesity programme and SURPASS is the diabetes one. The figure you quoted belongs to the other one.

comment on [Question] how do you actually verify appetite in c/glp1science · 234 points · 2 years ago

GIP receptor biology is genuinely unsettled — there is a live argument about agonism versus antagonism at the receptor — and the clinical results are robust regardless, which is an uncomfortable and interesting position.

a warm gel pack tells you the pack failed, not that the material did

Disagree with the panic framing. Lyophilised material tolerates ordinary transit conditions far better than these threads imply.
