Nothing here is medical advice, and the honest version of most of these posts is that they describe consequences rather than causes.

Right — mechanism gives you a direction. It never gives you an effect size and people use it as though it does.

Careful with naming anybody. The board rule holds even here, and especially here.

Receptor expression in a tissue is necessary but not sufficient for an effect. You also need the agonist to reach it at relevant exposure, and that is where a lot of confident mechanism talk falls down.

Stopped for financial reasons and regained most of it. Not a moral story, just a budget and a documented outcome.
