comment on three years of appetite threads, summarised so you do not have to read them in c/glp1science · 15 points · 1 year ago

Tried to build a mental model from mechanism alone and produced a confident prediction that the trial data flatly contradicted.

the peripheral and central stories are not in competition

comment on how much of what we believe about amylin actually comes from co-agonism threads in c/cagrilintide · 87 points · 1 year ago

Careful — that is a dosing intuition carried over from another board and there is no basis for it here.

Glucose-dependent insulin secretion is why hypoglycaemia risk is low as monotherapy: the effect scales with glycaemia rather than acting unconditionally.

Mine settled at week 56 and has not come back since, across two more steps.

comment on [Discussion] can we stop arguing about fatigue until somebody posts a number in c/sideeffects · 1 points · 2 years ago

Right, and the hair thing is worth repeating: it tracks the rate of loss rather than the compound, and it stops.

GIP receptor biology is genuinely unsettled — there is a live argument about agonism versus antagonism at the receptor — and the clinical results are robust regardless, which is an uncomfortable and interesting position.

Disagree. That is a preclinical finding in a rodent model and you are stating it as human pharmacology.

comment on am I the only one who found amylin harder than the injections in c/cagrilintide · 229 points · 2 years ago

Nothing containing this compound is approved as a standalone product, and research-use-only material is not approved for human use.

Correction: that is the combination programme, not the monotherapy readout. Different arms, different numbers.

What was the exposure in that experiment relative to therapeutic?

dose response is not linear and nobody should assume it is

Hair shedding after significant weight loss is a telogen effect that follows the rate of loss. It is not specific to these compounds and it resolves.

Constipation is mostly reduced intake plus reduced fluid, not a direct effect. That is why the boring fibre and water answer keeps working.
