comment on am I the only one who found tirzepatide harder than the injections in c/tirzepatide · 6 points · 2 years ago

Disagree with the conversion table. There is no validated equivalence between the two molecules and posting one as though there is does real damage.

comment on [Discussion] can we stop arguing about ATTAIN until somebody posts a number in c/orforglipron · 178 points · 2 years ago

The analytical point, which this board keeps getting wrong by importing habits from the peptide side.

For a peptide, purity is typically reported as area percent by HPLC with identity by mass spectrometry, and the impurity classes are things like deletion and oxidation products. For a small molecule the relevant impurities are synthetic intermediates, degradants and residual solvents, and identity is established differently.

So the certificate you would want looks different, the questions to ask are different, and a "purity" figure quoted here is not comparable to one quoted on the peptide boards. Anybody posting a result should say what method produced it, which is good practice everywhere and essential here.

a lot of the confident posting here is extrapolation

Yes. I stepped to 12.5 because the calendar said so, not because anything needed fixing, and I regretted it for a fortnight.

comment on genuine question about SURMOUNT that I am slightly embarrassed to ask in c/tirzepatide · 1 points · 2 years ago

Vials and pens carry the same molecule; what differs is fill volume, device tolerance and whether you are doing your own arithmetic. Neither is inherently more accurate.

Not convinced. That fee is the network fee at that hour, and it has nothing to do with the supplier.

phase 2 data only, and people quote it like it is a label

Adding the standing caveat: none of this is approved anywhere and research material is not for human use.

Is that the 48-week figure or an earlier readout?

Agreed, and the framing that helps is: this is a phase 2 compound with phase 3 running. Everything else is inference.

Sent a vial to Janoshik out of curiosity. Result was 98.4% against a claimed 97.5%, which is the first independent number I had seen for this compound anywhere.

Correction: TRIUMPH is the phase 3 programme. The number you are quoting is from the phase 2 readout, which is a different trial.

Careful — that would make individual moderation cases public by default, which is worse for the person complaining.

The ranking functions are published: the hot ordering combines score magnitude with age, and the controversial ordering weights total volume by the balance between up and down votes. Both are reproducible from the numbers on each submission.
