comment on SURMOUNT: what the trials say vs what this community says in c/tirzepatide · 68 points · 11 months ago
switching from sema is not a dose conversion, there is no clean equivalence
comment on appetite is the most under-discussed thing on this board in c/glp1science · -16 points · 11 months ago
Read the primary paper after arguing about the summary for a fortnight. The discussion section said almost the opposite of what the thread had concluded.
comment on contraception — 11 things I got wrong before I got it right in c/glp1women · 20 points · 11 months ago
Cosigning on fertility changing with weight loss. It catches people who were not expecting it at all.
comment on the half-life question that gets asked weekly, answered properly in c/glp1science · 0 points · 1 year ago
Correcting my own comment: I attributed that to the GIP arm and the paper attributes it to the GLP-1 arm.
comment on the half-life question that gets asked weekly, answered properly in c/glp1science · 38 points · 1 year ago
Is that from a human study or a preclinical model?
comment on someone explain reflux to me like I have not read a paper in years in c/sideeffects · 123 points · 1 year ago
the first fortnight after a step is the worst of it, then it settles
comment on someone explain reflux to me like I have not read a paper in years in c/sideeffects · 95 points · 1 year ago
Held the dose for an extra six weeks instead of stepping.
Disagreeing with this one line: that symptom is not on the wait-and-see list and should not be posted as though it is.
comment on what changed for me between month 11 and month 20 in c/t2dglp1 · 366 points · 1 year ago
Variability dropped before the average did, which was visible on the sensor and invisible on the lab result.
comment on [PSA] gallbladder is not what most of this community thinks it is in c/sideeffects · 48 points · 1 year ago
Agreed. Slowing down at meals did more for my constipation than everything I bought to fix it, which was embarrassing to discover.
comment on [PSA] gallbladder is not what most of this community thinks it is in c/sideeffects · 1 points · 1 year ago
holding a dose an extra month is a legitimate answer to bad tolerance
comment on [Question] is 93.7% actually fine or am I being sold a rounding error in c/tirzepatide · 1 points · 1 year ago
I would separate the two claims. That the GIP arm exists is uncontroversial; that it explains your fatigue pattern is a guess.
comment on the sulphur burps thing finally clicked for me and I want to write it down in c/sideeffects · 64 points · 1 year ago
Nobody warned me about early fullness and I would have liked the warning. It was not dangerous, it was just deeply unpleasant and completely unexpected.
comment on [Discussion] perimenopause is doing more work than we give it credit for in c/glp1women · 110 points · 1 year ago
Went looking for trial data on the specific question asked here and found the population had not been enrolled.
comment on [PSA] sulphur burps are not what most of this community thinks it is in c/sideeffects · 369 points · 1 year ago
A symptom that begins the day after the shot and fades by day four tracks the exposure curve. One that appears at random in week 76 probably does not, and that is worth saying to a clinician rather than to us.
comment on three years of appetite threads, summarised so you do not have to read them in c/glp1science · 5 points · 1 year ago
Was completely wrong about the gastric emptying story in a thread here two years ago. Someone corrected me with a citation and I have not made that mistake since.
comment on nobody warned me about sulphur burps and I would have liked the warning in c/sideeffects · 271 points · 1 year ago
reflux on day two and gone by day four is the standard shape
comment on [PSA] T2D is not what most of this community thinks it is in c/t2dglp1 · 48 points · 1 year ago
Kept fingersticks alongside the sensor for two weeks to sanity-check it. Worth doing once.
comment on the nausea thing finally clicked for me and I want to write it down in c/sideeffects · 20 points · 1 year ago
if it is severe or persistent that is a clinician question, not a board question
comment on PCOS — my 15-week log, condensed into one table in c/glp1women · 1 points · 1 year ago
I would not read one person’s cycle-linked experience as a general pattern. The variation here is enormous.
comment on how much of what we believe about dual agonist actually comes from GIP threads in c/tirzepatide · 0 points · 1 year ago
Saving this one.
kaia_wojcik is right that the two molecules are not interchangeable. There is no published conversion and the ones circulating are invented.
comment on [Meta] the T2D rule is doing its job and people should stop complaining in c/t2dglp1 · 1 points · 1 year ago
Cosigning that postprandial excursions improve first and the fasting number is the laggard.
comment on [Meta] the T2D rule is doing its job and people should stop complaining in c/t2dglp1 · 0 points · 1 year ago
Incretin-based agents stimulate insulin secretion in a glucose-dependent manner, which is why hypoglycaemia risk with them alone is low. Risk rises with agents that act independently of glycaemia.
comment on [Meta] the heart rate rule is doing its job and people should stop complaining in c/sideeffects · 7 points · 1 year ago
Has anything else changed in the same window?
comment on appetite — 15 things I got wrong before I got it right in c/glp1science · 1 points · 1 year ago
Long half-life in this class comes from structural modification that promotes albumin binding and resists enzymatic degradation. Weekly dosing is a consequence of the molecule, not a convenience decision.
comment on 7 months in and cycle is still the thing I get wrong in c/glp1women · 292 points · 1 year ago
I would not read one person’s cycle-linked experience as a general pattern. The variation here is enormous.
comment on unpopular opinion: most of what gets said here about CGM is guesswork in c/t2dglp1 · 9 points · 1 year ago
Same. The first day of a new sensor is unreliable and people rebuild their whole week around it.
comment on [Discussion] appetite is doing more work than we give it credit for in c/glp1science · 0 points · 1 year ago
I would not read that in vitro number across to a person. The conditions are nothing like physiological.
comment on [Meta] the cycle rule is doing its job and people should stop complaining in c/glp1women · 1 points · 2 years ago
say what you are actually asking, the vocabulary here is imprecise
comment on help me understand half-life, I have read the wiki twice in c/glp1science · 136 points · 2 years ago
That study was in a rodent model. Worth stating, since the thread has been reading it as human data.
comment on does half-life actually matter or is it forum lore at this point in c/glp1science · -4 points · 2 years ago
Cosigning on GIP. The genuinely interesting thing is that the biology is not settled and the clinical result is nonetheless robust.
comment on three years of receptor threads, summarised so you do not have to read them in c/glp1science · 75 points · 2 years ago
Is there any human data on that mechanism yet?
comment on how much of what we believe about heart rate actually comes from reflux threads in c/sideeffects · 188 points · 2 years ago
early fullness for the first eight days after each step, then nothing, every single time. Predictable enough that I started planning my week around it.
comment on someone explain incretin to me like I have not read a paper in years in c/glp1science · 39 points · 2 years ago
Started reading limitations sections first.
This is the concept everything else on this board is downstream of.
comment on genuine question about reflux that I am slightly embarrassed to ask in c/sideeffects · 247 points · 2 years ago
Mine settled at week 25 and has not come back since, across two more steps.
comment on [PSA] constipation is not what most of this community thinks it is in c/sideeffects · 181 points · 2 years ago
Severe or persistent abdominal pain sits outside the pattern this board can usefully discuss. The honest answer there is that it needs someone who can examine you.
comment on small win: fatigue stopped being a problem at week 82 in c/sideeffects · 674 points · 2 years ago
Same pattern here. It settled at a stable dose without me doing anything clever.
comment on does gastric emptying actually matter or is it forum lore at this point in c/glp1science · 712 points · 2 years ago
Tolerance to the gastric effect develops with continued exposure while the appetite effect largely persists. That single fact explains most of the "it settles but it still works" pattern the side-effect board reports.
comment on [Question] tirzepatide — what am I missing here in c/tirzepatide · -38 points · 2 years ago
Push back: "gentler than sema" is a population statement. Plenty of people on this board have the opposite experience and they are not doing it wrong.
comment on someone explain nausea to me like I have not read a paper in years in c/sideeffects · 1.7k points · 2 years ago
Constipation is mostly reduced intake plus reduced fluid, not a direct effect. That is why the boring fibre and water answer keeps working.
comment on how much of what we believe about sulphur burps actually comes from fatigue threads in c/sideeffects · 199 points · 2 years ago
nobody warns you about how much of this is timing rather than dose
