Weight change and appetite change are different endpoints on different timescales. Conflating them is where most of the confusion on this board starts.

System suitability — repeat injections, tailing factor, plate count, RSD on area — is what tells you the instrument was fit for the measurement that day. Without it, the purity figure is unanchored.

comment on dose history is the most under-discussed thing on this board in c/dosinglogs · 29 points · 1 months ago

Right, and the payoff arrives around month six when a pattern becomes visible that nobody could hold in their head.

comment on what would you tell week-1 you about 2.4mg in c/semaglutide · 11 points · 2 months ago

That reads like a titration plan and this board cannot give you one. Somebody who actually knows your history has to.

comment on why does nobody talk about BPC-157 in c/bpc157 · 28 points · 2 months ago

Assumed regulatory status was static and it was not. Checked again this year and the answer had changed where I live.

comment on is it normal to get nausea at week 48 in c/dosinglogs · 60 points · 2 months ago

Left up.

dose_diary_dana is right about blanks. Silent gaps make the series lie.

UV response is not uniform across species. At 214nm you are looking at the peptide bond, which is why it is the working wavelength for this class; at 280nm you only see tryptophan, tyrosine and phenylalanine.

Purity by area percent tells you the sample is homogeneous.

Adding the standing caveat — research material is not approved for human use, wherever you are.

comment on [Discussion] we are measuring weekly update at the wrong time and calling it noise in c/dosinglogs · 2 points · 3 months ago

Added a sleep column late and immediately understood two of my worst weeks.

Agreed — and the payoff genuinely arrives in month six, not week two.

staying at 0.5mg for an extra month is a legitimate plan

That figure cannot be quoted to two decimals off that baseline. The precision is not in the data.

Gave the same trace to two people I trust and got two integrations about a point apart. That was the most educational afternoon I have had here.

This. Mass spectrometry for identity, not just an area percent, because a pure something-else is still something else.

comment on someone explain gradient to me like I have not read a paper in years in c/hplc · 106 points · 4 months ago

Carryover from a previous high-concentration injection looks exactly like a small impurity. The blank injection is four minutes and it removes the ambiguity entirely.

permalink

comment on titration is the most under-discussed thing on this board in c/semaglutide · 67 points · 6 months ago

Delayed gastric emptying is most pronounced early and attenuates with continued dosing, which is why the early fullness usually fades at a stable dose.

permalink

comment on [Question] gradient — what am I missing here in c/hplc · 1 points · 7 months ago

Resolution between two peaks depends on retention, selectivity and efficiency. A shallower gradient buys retention and usually resolution, at the cost of peak width and run time.

Right, and clearance time is not carrier time. Blaming the shipper for a customs queue is a category error.

Sceptical of the mechanism you are proposing. Gastric emptying slows, it does not stop, and the timings you describe do not follow from it.
