comment on [Sceptic] every new three-letter analog needs a data table before a hype thread in c/researchpeptides · 1 points · 18 hr ago

identity before purity, every time, for anything unusual

net peptide content is where the surprises live

comment on [Method] the gradient everyone uses for sema and why it under-resolves the des-amido in c/hplc · 1 points · 3 days ago

Small fix — 214nm, not 210. It matters for the comparison you are making with the other run.

Correction: that is a tailing factor, not a plate count. Different diagnostics for different problems.

Net peptide content on the certificate was well below the purity figure and both were honest. Chemistry, not deception.

Disagree — that name refers to at least two different molecules depending on who is selling it. The thread needs to be specific.

Nothing containing this compound is approved by any regulator, and research-use-only material is not approved for human use.

This is the fact the whole board hangs on. Small molecule, not peptide.

Yes — system suitability first. Without it the number is an assertion about the instrument, not the sample.

Bought small on anything without a testing history here. That rule has never once cost me anything.

comment on research peptides — 22 things I got wrong before I got it right in c/researchpeptides · 11 points · 2 months ago

Purity methods differ by compound class. A gradient and wavelength developed for one peptide will not necessarily resolve the relevant impurities of another.

a blank injection between samples costs four minutes and settles most arguments

That is area percent, not mass percent. The trace cannot give you the second one.

comment on [Discussion] can we stop arguing about method development until somebody posts a number in c/hplc · 119 points · 3 months ago

Careful — you are treating retention time as identity. Two things can co-elute and the trace will not tell you.

comment on three years of chromatogram threads, summarised so you do not have to read them in c/hplc · 4 points · 3 months ago

Mass spectrometry answers identity. UV purity answers relative quantity under the run conditions. A document with one and not the other is answering half the question, and the half it answers should be stated.

you cannot report to two decimals off that baseline

permalink

Agreed — identity first for anything off the well-travelled list. The purity number is meaningless until you know what it is pure of.

Independent result added to the log — there are very few on that compound.

Disagreeing with this bit: handling for one compound does not transfer to another by default.

Appetite change and weight change are different measurements on different timescales.

Agreed, and for anyone finding this later: that is the normal experience, not a warning sign.

Cosigning the wavelength point. Half the disagreements in this board are two people looking at different detectors.

That figure cannot be quoted to two decimals off that baseline. The precision is not in the data.

Do you have the mass, or only the UV trace?

permalink

comment on what would you tell week-1 you about non-peptide in c/orforglipron · 7 points · 12 months ago

Careful — purity methods for peptides do not read across to a small molecule and the numbers are not equivalent.

Went looking for independent testing on this and found essentially nothing, which was clarifying.

Right. And a blank between injections settles the carryover argument before it starts.

I would not treat catalogue breadth as a quality signal. It is a business decision, not an analytical one.
