Phase 2 to phase 3 has historically not been kind to effect sizes in this class. Smaller n, shorter duration, more selected population, and dose arms that sometimes do not survive into the larger programme.
None of that means the number is wrong. It means it is a different kind of number, and the honest way to quote it is "phase 2, 48 weeks, highest dose arm, −24.2%", which is longer and less exciting and correct.

SUSTAIN 6 and the STEP program are both once-weekly designs, pretty sure none of the major published trials tested a split schedule directly, so anyone hunting for a hidden RCT on this is going to come up empty.

worth flagging, none of the major trials (STEP program, SURMOUNT-1, SUSTAIN 6) actually tested a deliberate dose-reduction-at-maintenance protocol, they were all escalation-to-target designs, so this whole "floor hunting" practice is genuinely undocumented territory pharmacologically, not a formally studied strategy. edit: originally wrote "SURMOUNT-4" here which is about withdrawal, not maintenance dose-reduction, wrong citation, fixed.

On the thin testing history in this board, and what to do about it.
Most of the compounds discussed here have a handful of independent results across the entire site, and several have none. That is a fact about how few members have paid for testing, not a judgement about the material.
What follows: buy small, test identity first for anything unusual, and post the result even when it is unremarkable. The vendor source pages are built from exactly these submissions, and the compounds with the thinnest coverage are the ones where one more entry changes the picture most. Research material is not approved for human use, so what this board can usefully build is an analytical record.

Why purity and net content diverge, and why that is usually honest.
Purity by area percent is a relative measure of what the detector saw. Net peptide content subtracts counterion, water and residual solvent from the powder mass. For some compounds in this catalogue the gap between the two is large.
A vial can therefore be 99% pure and contain considerably less peptide by mass than the label weight suggests, with nothing wrong anywhere. The problem is not the chemistry; it is that people compare a purity figure from one supplier with a net content figure from another and conclude something about both. Ask which number you are being shown, every time.

Reading a trace posted here, in the order I actually look at things.
Axes first — if the wavelength and the time axis are not labelled, I stop. Then the baseline: where has it been drawn, and does the drawing absorb anything. Then peak shape: fronting, tailing, a shoulder that never resolves. Then the blank, if one was run, for carryover.
Only after all of that do I look at the percentage, and by then I usually know how much weight it deserves. The number is the last thing on the page and the first thing everybody argues about, which is exactly backwards.

The analytical point, which this board keeps getting wrong by importing habits from the peptide side.
For a peptide, purity is typically reported as area percent by HPLC with identity by mass spectrometry, and the impurity classes are things like deletion and oxidation products. For a small molecule the relevant impurities are synthetic intermediates, degradants and residual solvents, and identity is established differently.
So the certificate you would want looks different, the questions to ask are different, and a "purity" figure quoted here is not comparable to one quoted on the peptide boards. Anybody posting a result should say what method produced it, which is good practice everywhere and essential here.

Once in solution, the relevant risks are hydrolysis, aggregation and, for some sequences, oxidation. Refrigeration slows all three; freezing a solution introduces a separate risk at the ice interface.

Sent the same vial to PeptideMeter and VendorInvestigate. 98.8% against a claimed 98.5%. The difference was the gradient, not the material.

What a result posted here is actually worth, and how to make yours worth more.
A single figure with no batch identifier is about a vial nobody can locate. Add the batch and it becomes about a lot. Add the claimed figure from the certificate and it becomes a comparison. Add the service and the date and it becomes reproducible. Say that you paid for it yourself and it becomes independent.
Five extra fields, thirty seconds of typing, and the difference between a comment and a row in the log that will still be useful in three years.
