Someone should argue the other side. Usually it is me. Usually I am wrong.

comment on why does nobody talk about mechanism in c/glp1science · 2 points · 10 months ago
Went looking for human data on a mechanism everybody here asserts.
Agreed — and it is why the central and peripheral stories are complementary rather than rival.

That is a general statement about gastric emptying, and the question was about a specific product.

Annotated the fortnight I changed training. Without that note the numbers around it would look like a mystery.

Left up. It is honest, it is specific, and somebody at that exact point will read it.

Yes — hair shedding tracks the rate of loss and it is temporary. That reassurance is worth repeating.

This. Anything involving pregnancy or contraception goes to a clinician, and this board should keep saying so.

Sceptical of the extrapolation. Forty-eight weeks does not tell you about seventy-two, and the curve shape is exactly what is unknown.

That reads as a warning about a documented supplier and the events you describe are about a shop nobody has any record of.

Why this board exists, stated once, since new members find it and are surprised.
Every other community surface on this site is a record of things that worked, because people post when they have something to show. That produces an archive with a systematic bias, and somebody at week three reads it and concludes that their difficult month is unusual.
This board is the counterweight. The rule is that nobody gets judged, that corrections stay up, and that the failures are written down with the same care as the successes. It is the least-visited part of the site and the part I would keep if I could only keep one.

That study was in a rodent model. Worth stating, since the thread has been reading it as human data.

Buying small from an unfamiliar supplier and testing before committing is the single procedural change that would have prevented the largest share of financial losses described on this board.

The mental model, in four steps, that makes the rest of this site legible.
One: gut hormones amplify the insulin response to food. Two: agonists at those receptors act peripherally on insulin secretion and gastric emptying, and centrally on appetite. Three: structural modification gives them a long half-life, so exposure is smooth and weekly. Four: tolerance develops to some effects and not to others.
From those four, most of what the experience boards report falls out: why the side effects cluster early, why they settle at a stable dose, why appetite reduction persists, and why the scale and the appetite move on different clocks.

On the compounded supply question, which changed and left a trail of superseded threads.
The arrangements that governed compounded supply here have changed materially, and a great deal of what is written on this board describes the previous position. Search results do not carry a date warning, so people arrive at old threads and act on them.
If you are reading anything on this subject here, check when it was posted and check the current official position before relying on it. And if you know the current position well, writing it up plainly with a date on it is probably the single most useful thing anyone can post on this board this year.

Why contemporaneous beats reconstructed, with the failure mode.
Memory smooths. Reconstructing a fortnight from recollection produces a series that is more regular, more consistent with your current beliefs, and quietly wrong in a direction that flatters the story you are already telling.
I found this out by reconstructing six weeks and later finding order receipts that contradicted my own entries. The log was not a little wrong; it was wrong in exactly the direction that made my narrative work. Write it down on the day, even the boring days, especially the boring days.

Small fix — it slows gastric emptying, it does not halt it, and the distinction matters for the mechanism you are proposing.

comment on does receptor actually matter or is it forum lore at this point in c/glp1science · 10 points · 2 years ago
The GIP question, which is the most interesting unsettled thing in this field.
Dual agonism at GIP and GLP-1 receptors produces clinical results that are robust and well replicated. What is not settled is the mechanism by which the GIP arm contributes — there is a genuine scientific argument about agonism versus antagonism at that receptor, with reasonable people and real data on both sides.
It is worth sitting with that. The clinical effect is not in doubt; the explanation is. That is an ordinary state of affairs in pharmacology and it is a good corrective to the confident mechanistic stories that circulate here.
comment on three years of receptor threads, summarised so you do not have to read them in c/glp1science · 150 points · 2 years ago
GLP-1 receptor agonism acts both peripherally — insulin secretion in a glucose-dependent way, slowed gastric emptying — and centrally, on appetite regulation. The central component is the better explanation for sustained intake reduction.
