the trend line matters more than any single value

one measurement is a point, two is a line, three is a trend

Cardiovascular outcome trials are powered for events, not for weight, and are typically run in a different population with different inclusion criteria. Reading a weight number out of one is reading a secondary endpoint.

The rules where I live changed between two orders six months apart. Nothing was announced; I found out by reading.

Careful with the rankings. A phase 2 result and a phase 3 result are not on the same evidential footing and cannot be listed in one table.

The phase 2 readout ran to 48 weeks in a few hundred participants with a slow escalation. Both the size and the duration matter when people quote the headline number.

Does constipation follow the day after the shot, or is it spread across the week?

Kept every document including the dull ones.

Agreed — read the notice before deciding what has happened. Most of the panic here is misclassification.

staying at 1.7mg for an extra month is a legitimate plan

Minor fix — the half-life is about a week, not about a day. The rest of your point stands.

Are you measuring anything other than weight? Waist is usually the one that keeps moving.

Same experience with the appetite effect being flatter across the week rather than front-loaded.

Do you have a baseline from before you started?

Small fix — a reference interval is not a treatment target, and the post above uses them interchangeably.

Not convinced. You changed the dose and the week at the same time, so you cannot attribute the change to either.

Substantial weight loss independently moves lipids, liver enzymes and several other markers. Attributing a change to a compound while losing weight is confounded by design.

comment on Mounjaro: the version I wish someone had shown me in week 1 in c/tirzepatide · -18 points · 4 months ago

Switching from semaglutide, since three people asked in this thread alone.

There is no published dose equivalence between the two. The conversion tables that circulate are somebody’s arithmetic, not data. What people report here is that the first month after a switch is often flat, that the appetite effect feels differently shaped rather than simply stronger, and that starting at the bottom of the ladder again is the common approach.

None of that is a recommendation. It is what the threads say, and the threads are not a clinic.

Small fix — those rules apply to commercial import. Personal import is governed separately and more narrowly.

Research-use-only material is not approved for human use. Anything in this thread about how a research vial "should" titrate is a discussion about chemistry, not a plan.

With a week-long half-life, exposure declines over several weeks after the last dose rather than stopping with it. The lag is why the first fortnight is uninformative.

My ferritin moved and I spent a fortnight convinced I knew why. The repeat six weeks later was back in range and I had changed nothing.

That is the 68-week readout, not the 72-week one. Different trial, different duration.

Correction: TRIUMPH is the phase 3 programme. The number you are quoting is from the phase 2 readout, which is a different trial.

Interval stretching lowers the trough concentration while keeping the dose; dose reduction lowers the level while keeping the rhythm. They are different interventions with different subjective profiles.

2.5mg is a four-week starting dose, not a maintenance dose. Worth being precise since people search these threads.

Correcting myself from earlier in the thread: I said week 10 and it was week 18. Same shape, wrong number.
