comment on [Results] 14 weeks on 5mg, full numbers, ask me anything boring in c/glp1muscle · 19 points · 10 months ago

Strength retention is an imperfect but free longitudinal proxy: it is affected by neural factors and by being in a deficit, so it tracks tissue change loosely rather than exactly.

comment on [Vendor] TFC order #8 — Denmark, 12 days, no drama, receipts inside in c/cryptopay · 27 points · 10 months ago

Cosigning on keeping the hash. It is the only receipt in existence and people screenshot their balance instead.

permalink

comment on [Question] Canada — has anyone got a straight answer on escrow in c/cryptopay · 10 points · 11 months ago

Yes — the same token symbol on two chains catches people constantly and it is unrecoverable.

comment on someone explain chargeback to me like I have not read a paper in years in c/cryptopay · 8 points · 12 months ago

Asked which networks SGN accepted before ordering and picked the cheapest one at that hour. Saved more than the difference in price.

dose response is not linear and nobody should assume it is

permalink

comment on how much of what we believe about mechanism actually comes from appetite threads in c/glp1science · 0 points · 1 year ago

Not convinced by the linearity assumption. Dose-response in this class is not linear and the trials show it.

comment on the lean mass thing finally clicked for me and I want to write it down in c/glp1muscle · 101 points · 1 year ago

Right — strength retention is a free proxy and it tracks better than most people expect.

permalink

comment on [Question] how do you actually verify DEXA in c/glp1muscle · 15 points · 1 year ago

DEXA has a real error margin and people quote it to one decimal

permalink

comment on genuine question about dual agonist that I am slightly embarrassed to ask in c/survodutide · 16 points · 1 year ago

That figure is from the obesity programme, and this thread is about the hepatic one.

comment on does protein actually matter or is it forum lore at this point in c/glp1muscle · 1 points · 1 year ago

Have the lifts held, dropped, or moved around?

permalink

comment on hepatic fat — 19 things I got wrong before I got it right in c/survodutide · 104 points · 1 year ago

It is a dual agonist at the GLP-1 and glucagon receptors. The glucagon arm is associated with increased energy expenditure and with effects on hepatic fat, which is why the development programme is oriented towards liver disease.

comment on [Meta] the confirmations rule is doing its job and people should stop complaining in c/cryptopay · 172 points · 1 year ago

I would not treat a delayed confirmation as evidence of anything at all. Chains have busy hours.

permalink

comment on help me understand DHL, I have read the wiki twice in c/intlshipping · 22 points · 1 year ago

Yes — median rather than best case. The fastest anybody ever got is not a plan.

permalink

comment on [Meta] proposal — a flair for half-life posts in c/glp1science · 373 points · 1 year ago

Receptor expression in a tissue is necessary but not sufficient for an effect. You also need the agonist to reach it at relevant exposure, and that is where a lot of confident mechanism talk falls down.

Pasted rather than typed the address, then checked the first and last six characters. Habit now.

Posted my first lane report here without the month and got asked for it. Fair, and the table is better for the fussiness.

The transaction hash is the canonical receipt: it is public, verifiable by either party, and it is what a support query should quote.

permalink

Agreed that quote expiry is ordinary. Rates move and a time-limited quote is not a pressure tactic on its own.

Agreed — and the hash is the receipt.

Adding the cheap insurance — test amount first on anything large.

permalink

comment on SURPASS — 9 things I got wrong before I got it right in c/trialwatch · 469 points · 2 years ago

The registered protocol is public. Comparing the registered primary endpoint with the reported one is a two-minute check and it is how outcome switching gets caught.

permalink

comment on why does nobody talk about MASH in c/survodutide · 50 points · 2 years ago

Yes — nothing containing this is approved anywhere, which is the first fact and usually the last one mentioned.

permalink

comment on why does nobody talk about MASH in c/survodutide · 343 points · 2 years ago

Nothing containing this compound is approved anywhere. Research-use-only material is not approved for human use.

That conflates receptor affinity with clinical potency. They are related and they are not the same thing.

permalink

comment on reship — 8 things I got wrong before I got it right in c/intlshipping · 1 points · 2 years ago

Total transit decomposes into first mile, international leg, clearance and last mile. Only the first two are meaningfully under the shipper’s control and most reported variance sits in the last two.

permalink

comment on reship — 8 things I got wrong before I got it right in c/intlshipping · 311 points · 2 years ago

the last mile is where most of the variance lives

permalink

comment on DHL is the most under-discussed thing on this board in c/intlshipping · 17 points · 2 years ago

Same. TFC quoted 6 days on my lane and it landed at 14, which is exactly what you want from a quote.

comment on [Discussion] incretin is doing more work than we give it credit for in c/glp1science · 48 points · 2 years ago

Long half-life in this class comes from structural modification that promotes albumin binding and resists enzymatic degradation. Weekly dosing is a consequence of the molecule, not a convenience decision.
