GIP/GLP-1 co-agonism is genuinely interesting pharmacology, separate from the weight story.

comparing reta phase 2 to sema phase 3 is comparing different things

Ignored something for four days that I should have asked about on day one. That is the only genuine regret in my whole run.

SURMOUNT-1 ran 72 weeks with the top arm around 21% mean body weight change.

Not sure about this bit. The GIP arm being real does not tell you that it is what caused your particular week.

Regulatory classification differs between jurisdictions and has changed in several of them recently. Any answer to a status question needs a place and a date attached.

Correction: those scans are from different machines, and the difference you are describing is within the measurement error.

comment on regain: what the trials say vs what this community says in c/maintenancephase · 52 points · 1 months ago

Withdrawal studies in this class consistently show substantial regain after discontinuation. That is a pharmacological finding about a chronic condition, not a statement about anyone’s discipline.

The nausea profile felt different rather than worse. Hard to describe and impossible to quantify, so take it as an impression.

Retitled to distinguish preclinical from clinical, which the original ran together.

comment on [Results] 48 weeks, 18kg, and DEXA was the hard part in c/glp1muscle · 1 points · 2 months ago

DEXA has a meaningful coefficient of variation, and it differs between machines and protocols. Comparing scans from different clinics adds a systematic offset that is invisible on the report.

Left up, flair changed to Discussion. It is an opinion post and that is fine as long as it is labelled.

dose response is not linear and nobody should assume it is

Sceptical of the extrapolation. Forty-eight weeks does not tell you about seventy-two, and the curve shape is exactly what is unknown.

Not convinced. Interval stretching changes your exposure profile, not just the total, and the two are not equivalent.

It is a triple agonist at the GLP-1, GIP and glucagon receptors. The glucagon arm is the genuinely novel component and it is the one with the least human outcome data behind it.

Weigh monthly now. Best change I made in the whole run and it cost nothing.

Two years at maintenance. The logistics are the only remaining work — ordering, storing, remembering.

Retitled to name the endpoint. Histological and imaging results are not interchangeable and the original title implied they were.

Small fix — it slows gastric emptying, it does not halt it, and the distinction matters for the mechanism you are proposing.

Correcting my own comment: I attributed that to the GIP arm and the paper attributes it to the GLP-1 arm.

Does the effect persist with continued dosing or does tolerance develop?

Interval stretching changes the exposure profile: with a week-long half-life, moving to ten or twelve days lowers trough concentration more than it lo

This reframing is the most useful thing on the board for anyone approaching this phase.

That reads as a warning about a documented supplier and the events you describe are about a shop nobody has any record of.

Agreed — and being harder on yourself than you would be on somebody else posting the same thing is the pattern here.

None of this is a taper plan. What dose holds for you is a question for somebody with your history in front of them.
