Purity by area percent tells you the sample is homogeneous. Mass spectrometry tells you what the homogeneous thing is. For a short peptide, the second question is the one worth paying for.

Both, but not in the direction I expected.
I buy from fewer places and I test more of what I buy. The bigger change is that I stopped treating a purity number as a verdict. Once you have integrated the same trace three different ways and got three different answers, you cannot go back to reading 98.6 as a fact about the world.
Which sounds like nihilism and is not. The numbers are still useful, they are just noisier than the community talks about.

yes and neither of those two vials is the bad case. i have seen a 97.9% that was basically one enormous shoulder and that one worried me a lot more than a 96 with a clean profile.

Why step therapy denials feel unfair and are nonetheless beatable.
The requirement is that documented trials of preferred alternatives exist. It is a record-keeping standard, not a clinical judgement about you, which is why arguing the clinical merits rarely moves it and producing dates and durations often does.
What to assemble: what was tried, at what dose, for how long, and what the documented outcome was. A one-page table with dates beats three pages of prose every time. Where a trial is contraindicated rather than simply unsuccessful, that needs to be stated explicitly by the prescriber in those terms.

A shoulder that does not baseline-resolve cannot be quantified honestly. You can report it as an unresolved shoulder, which is useful information, or you can develop the method until it resolves.

Why two honest labs report different numbers on the same vial.
Start with the gradient. A shallower slope holds compounds on the column longer and usually separates close-eluting species better. A steeper one gets you a faster run and a fatter peak. If a related substance elutes near the main peak, one method resolves it and reports it separately, the other absorbs part of it into the main peak.
Then integration. Where the baseline is drawn under a shoulder is a decision made by a person or by a piece of software configured by a person. It moves the number.
A point or two of spread between services on this assay is ordinary. Treating one lab as ground truth is how people end up in arguments with suppliers that neither side can win.

The three questions to ask before ordering anything from the long tail of a catalogue.
What exactly is it — catalogue number or sequence, not a common name, because several common names map to more than one molecule. How was identity established — mass against theoretical, ideally, rather than a bare purity figure. What are the storage conditions, stated specifically, with a state and a duration attached.
Then two commercial questions that save time: is this a stocked line or made to order, and what is the lead time in writing. All five are one email, and the shape of the reply tells you a great deal before any material moves.

Gel packs are designed for a duty cycle. A liquid pack at the end of a longer-than-expected transit is a pack behaving as specified, not evidence of an excursion in the vial.
