comment on [Baseline] get these before you start, you cannot go back for them later in c/bloodwork · 23 points · 1 days ago

Push back: a value just outside a reference interval is a common finding in healthy people. It is a reason to repeat.

fair. writing it up properly.

while we are here: can we get a flair between Question and Discussion for posts that are really "argue with me"

comment on [Meta] should this community rename "maintenance phase" to something that doesn’t sound like an afterthought in c/maintenancephase · 3 points · 3 days ago

tangential but a "Floor Hunting" flair might genuinely be worth adding given how often that specific topic comes up here

correctly flaired Milestone too, small thing but I appreciate it

comment on [Meta] should titration questions get a megathread or is that overkill in c/newbiequestions · 11 points · 3 days ago

slightly off topic but half these threads are also missing the Titration flair when they'd qualify, that would help sorting on its own

comment on [Question] small molecule means purity testing is a different problem, right in c/orforglipron · 5 points · 3 days ago

Push back: daily dosing is not automatically better adherence. It is a different failure mode, not a solved problem.

comment on [Paper] receptor distribution and why the GI effects were predictable in c/glp1science · 33 points · 6 days ago

Long half-life in this class comes from structural modification that promotes albumin binding and resists enzymatic degradation. Weekly dosing is a consequence of the molecule, not a convenience decision.

What did the repeat show?

a non-peptide agonist does not degrade the way a peptide does

comment on week 62 check-in — 20kg down, muscle cramps manageable, one thing confusing me in c/dosinglogs · 1 points · 26 days ago

Careful — posting a log as a schedule for other people to follow is not what this board is for.

comment on log — my 20-week log, condensed into one table in c/dosinglogs · 2 points · 1 months ago

A log is only interpretable if the conditions are consistent.

ireland_drugs_pay is right about blanks. Silent gaps make the series lie.

comment on unpopular opinion: most of what gets said here about log is guesswork in c/dosinglogs · 32 points · 1 months ago

Not convinced by the retrospective reconstruction. Memory smooths things and the smoothing is systematic.

comment on [Discussion] we are measuring A1c at the wrong time and calling it noise in c/bloodwork · 2 points · 1 months ago

I would not draw a line through two points, especially when the second was taken at a different time of day.

Daily dosing, so what does the exposure profile look like across the day?

permalink

comment on reading ATTAIN threads from 2024 and half of it aged badly in c/orforglipron · 17 points · 1 months ago

the tolerability profile reads broadly similar to the class

Phase 2 or phase 3?

comment on reading ATTAIN threads from 2024 and half of it aged badly in c/orforglipron · 16 points · 1 months ago

Went looking for independent testing on this and found essentially nothing, which was clarifying.

Small fix — the two programme names got swapped upthread and it changes which population the figure came from.

Was completely wrong about the gastric emptying story in a thread here two years ago. Someone corrected me with a citation and I have not made that mistake since.

The confounding problem, stated plainly, because this board keeps stepping on it.

Substantial weight loss moves lipids, liver enzymes, insulin sensitivity markers and several others in its own right. If you are losing weight while taking something, any change in those markers has at least two candidate explanations and you cannot separate them from your own panel.

What you can do is standardise, take a baseline, keep the series long, and be honest in your posts about what is and is not attributable. The threads that say "this compound did X to my hs-CRP" almost never have the design to support the claim, mine included.

comment on [Discussion] can we stop arguing about half-life until somebody posts a number in c/glp1science · 29 points · 5 months ago

Do you have the paper, or a summary of it?

annika_fonseca is right that mechanism gives direction and not magnitude. Worth pinning.

I would not draw a trend line through that. Two points and a gap is not a series.

permalink

This. Reference ranges are population intervals, not targets, and conflating them causes a lot of unnecessary alarm here.

comment on [PSA] struggle is not what most of this community thinks it is in c/progresspics · 23 points · 6 months ago

Rate of change is not a virtue. Slower trajectories are as durable as fast ones and are considerably better represented on this board than the front page suggests.

Yes — log what happened rather than what you intended. My retrospective entries were quietly wrong every time.

Why this board is text only, since new members ask every week.

receptor_bias_rick is right that the clothes tell you before the scale does.

albumin binding is most of the half-life story

That conflates receptor affinity with clinical potency. They are related and they are not the same thing.

permalink

comment on my triglycerides moved and I cannot work out whether weekly update is why in c/dosinglogs · 377 points · 9 months ago

The four columns that survive, after several people here have tried and abandoned much more elaborate systems.

Date. Dose. One number measured under consistent conditions. A short note when anything unusual happened. That is it. Anything more and you will stop, and a log you stop keeping is worth less than a crude one you maintain.

Add a waist measurement monthly if you can be bothered, and a sleep column if you want the one variable that explains the most confusing weeks. Everything else can be reconstructed from those, and nothing can be reconstructed from a log you abandoned in week nine.

permalink

comment on weight: the version I wish someone had shown me in week 1 in c/dosinglogs · 37 points · 9 months ago

Attribution requires that one thing change at a time. A log with dose, training and intake all moving in the same fortnight cannot assign anything to any of them.

Why this board is text only, since new members ask every week.

Agreed — measurements over photographs, every time, for anybody trying to actually track something.
